A Study of Some Blood Picture Parameters during Experimental Infection with Eimeria tenella in Local breeds and Foreign of Chicken

The study was conducted in Animal house of the department of Biology, College of
Education, Al-Qadisiya University during the period from 1/9/2013 to 1/9/2014. The study
diagnosed the parasite of E.tenella in naturally infected chickens in poultry farms. E.tenella was
isolated from the infected cases in Vet. hospital and Vet. clinics.Then,the cases diagnosed initially
in the laboratory direct examining stool by ordinary microscope and the positive cases were
recorded and diagnosed by using the technique of Conventional Polymerase Chain
Reaction(PCR). There are 315 cases of infected chicken which are examined and the results
showed that (186 out of 315) stool Samples,59%, The experimental infection for each experiment
was done in age 21 day with dosage of (5×104) grown oocysts/chicken for E.tenella in two types
of chickens(Ross and local breed) Also,two groups of uninfected chickens (control) are used for
the sake of comparison.
Hematological parameters in Ross chickens showed significant decrease (p<0.05) in fourth
week of infection), percentage of Packed cell volume (PCV)(18.47% in the fifth week of
infection) and Red blood cell (RBC) count was (1.58x106 in the fourth week of infection) . Also ,
the infection led to significant difference of white blood cell (WBC) count (30.19x103 in the
seventh week of infection) comparison of uninfected chickens of the control group. While The
hematological parameters in local breed showed significant decrease (p<0.05) in Hb
concentration and percentage of PCV (7.13mg/100ml , 21.26% respectively in the fifth week of
infection) , while the RBC count was (2.05 x 106 in the fourth week of infection and significant
increased (p<0.05) in WBC count was (30.63 x 106 the sixth week of infection) in comparison
with the chickens of the control group.

The results of this study were including detection of Entamoeba histolytica in an in patients infected infants with diarrhea in born and childrenʾs hospital in Al-Diwania province during the period from June until December 2014, the number of samples was 170 stool sample for age between one month-two years, these samples were examined by direct smear method by using physiological normal saline and Iodine solution, there 88 samples were positive in percentage %51.76 also the lower percentage was %36.17 recorded in age 1-6 months and the higher percentage was %61.76 in age (1-6)-two years, wherever the percentage of an infected male was closely which reach %53.16 ,%50.55 respectively. According to type of feeding, the higher percentage was %59.79 in an infants with Artificial feeding compare with the percentage %41.09 in an infants with breast feeding, wherever the percentage of infection with parasite was %57.32 in rural region ,its higher than the percentage %59.46 in urban area, also the results were showed the percentage of infection with this parasite was %44.32 by using Real-time polymerase chain reaction as confirmative test .

Identification of cryptosporidium parvum parasite oocyst using microscopic &PCR assay for detection GP900gene

The present study designed to diagnose the parasite Cryptosporidium parvum from patients with diarrhea arrivals to Maternity and Childhood Teaching Hospital and General Education Hospital in Al-Qadisiya Governorate , as well as to investigate and to emphasize the presence of virulence factor Glycoprotein 900 in parasite C. parvum using Conventional PCR Technique, and for these purposes one hundred feces samples were collected from children under 10 years old, for the period between 1 5 2013 - 1 11 2013.
The results show presence of Cryptosporidium parvum parasite oocyst in 28 positive samples distributed throughout the months of the study, and then, DNA was extracted from the positive samples and then after amplified using special designed for primers C. parvum virulence factor gene that called Glycoprotein 900 and the amplified DNA passed in electrophoresis apparatus for DNA. The results showed that the mentioned factor is present in all positive samples of C. parvum.
The highest percentage of infection with the parasite was in the age group (1-3 years) with (39.2%), followed by age group (less than one year) with (21.4%) and the lowest percentage of infection was in the age group (7-10 years) by 7.1%, whereas the percentages of infection with the parasite in the age groups (5-3) , (7-5) were 17.8% , 14.2%, respectively.
Also the results of the current study, showed that there are no significant differences according to sex .The number of infected males were about 15 with (53.5%) and the number of infected females were about 13 with (46.4%) , besides the results indicate that the highest percentage of infection was in rural areas rose (57.1%) and the lowest percentage of infection was in the urban (42.8 %).

Identification study of Giardialambliawhich associated with drinking water in some areas in AL-Diwaniacity by using flotation method and ConventionalReceved

The present study which including testing 140 samples of drinking water which taken from
seven area in AL-Diwanyiaregion by using the flotation
method and conventional-PCR for identified and diagnosed of G.lamblia
,the results of this study refers into the
percentage of drinking water contamination was %22.85.Also, the results were showing, the
higher percentage of contamination was recorded in drinking water in the AL-Sadder area
was %45 and the lowerpercentage was in AL-Tagia and AL-Hokum area which was %5 of
each one of them.
The results of statistical analysis ,showing there were significant differentiation in the
percentage of drinking water between areas in probabilitylevel 0.05.Also the results of
confirmative detection of Giardin gene by using conventional-PCR, showing the percentage
of contamination drinking water was %5.71.
